Smith MR et al. – Denosumab in men receiving androgen-deprivation therapy for prostate cancer
Methods- Double-bline, multicenter study
- 734 patients (in each group) were randomly assigned to received densoumab (60 mg sc q6 mos) or placebo
- At 24 mos, BMD of the lumbar spine increased by 5.6% in the denosumab group vs. a 1.0% loss in the placebo group
- Significant differences occurred as early as 1 mo and were sustained through 36 mos
- Denosumab tx increased BMD at the total hip, femoral neck, and distal 1/3 of the radius
- Patients txd with denosumab had a decreased incidence of new vertebral fractures at 36 mos
- Adverse events in the two groups were similar
