| It was previously shown that treatment of renal cell carcinoma with high energy UVC (3,2 J/cm2) increases the tumour uptake by dendritic cells and activates the genes involved in the IFN-alpha driven antigen processing machinery (Brusa et al. Vaccine, 2008), thus speedingup the recruitment of antitumor lymphocytes. The effect was accompanied by the release of HSP-70 and HMGB1, two powerful immunoadjuvants. The present paper by the same authors (Prostate 2009) goes deeper into this issue, by showing that both UVC and high (56? C), but not intermediate, temperature can promote these effects on a prostate cancer (PCa) cell line. This finding has important implications in the understanding of the mechanism of action of focal therapy. When appropriately delivered, heating or UVC can create in the patient an immune, long lasting protection against recurrence, through the production of a highly immunogenic source of tumor antigens for dendritic cell loading in vivo. In addition, the observation that only defined conditions produce this effect may help improving the standards of future focal therapy approaches. |
