Stankiewicz E et al. - The low level of p16INK4A expression and human papillomavirus detection suggests that penile verrucous carcinoma pathogenesis is unrelated to human papillomavirus infection and the oncogenes and tumour suppressor genes classically altered by virus infection.

The aetiology and pathogenesis of penile carcinoma in still somewhat contentious. In spite of the prevalence of HPV in the community, when compared with cervical cancer, penile malignancy is very rare. other factors are presumably involved in the evolution of these tumours. verrucous tumours are a rare subtype, and this study correlates the lack p16 expression and concurrent lack of high risk HPV in these cases. We therefore believe that different penile tumour subtypes have different modes of pathogenesis. The implications of this are that these rare tumours are unlikely be eradicated by HPV vaccination in the future. Allthough usually benign, it should be noted that these tumours can recur and progress locally.
