Genetic risk and protective factors for the idiopathic inflammatory myopathies

O’Hanlon TP et al. - This article summarizes the recent findings on genetic risk and protective factors that might help us in understanding of the pathogenesis and treatment of the idiopathic infl ammatory myopathies (IIM). The conclusions drawn from this overview are:
- IIM, a group of systemic autoimmune syndromes likely results from complex environmentalinteractions with polygenic risk and protective factors
- While some genetic susceptibility factors are common to all forms of myositis, others are uniqueto particular phenotypes
- These phenotypic variations in associated polymorphisms may reflect divergent pathogenicmechanisms and/or varied responses to unique environmental triggers in different groups of pts
- High throughput whole genome polymorphism scans, coupled with array-based differentialgene and protein expression analyses has revealed the complex physiologic pathways involved
- Coordinated international collaborations are needed to obtain the necessary numbers of patientsfor future genome-wide association studies
