Drug-induced autoimmunity
Current Opinion in Rheumatology, 08/17/09
Dedeoglu F et al. - Even though drug-induced autoimmunity (DIA) has been known for many years, the underlying mechanisms remain unclear. However, with recently described new and unexpected features, novel hypotheses have been proposed, that may lead to further research in understanding these mechanisms.
Methods- This review discuses the features of DIA, including both clinical and autoantibody profiles in addition to the potential chronicity of the syndrome
- Use of newer biologics, mostly directed towards immune system manipulation has widened the range of signs and symptoms of DIA, and the patterns of autoantibody profiles
- Rashes and visceral involvement are reported more often
- In addition, anti-double stranded antibody, which are usually seen with idiopathic SLE are appearing in place of antihistone antibodies
- Finally, some medications have been implicated in causing the very same entity, which they may be used to treat
- Progress in the field of pharmacogenetics and pharmacogenomics will help further our understanding of these and other adverse effects of medications
