| This paper provides an overview on the molecular pathology of localized cutaneous amyloidosis - an oftentimes itchy skin disorder. The emerging data implicate an abnormaity of cytokine signaling in the skin that involves various interleukin-6 family cytokine receptors, including interleukin-31 signaling. This is a recently discovered cytokine and its overexpression (or that of its receptor) may also be found in other itchy dermatoses such as nodular prurigo and atopic dermatitis. Moreover, blocking the cytokine with antibodies in a mouse model of atopic dermatitis can reduce scratching. Collectively, the work on localized cutaneous amyloidosis and other studies on interleukin-31 suggest that modulation of this cytokine might have a significant role to play in the management of itch. In the future we may see a new class of clinically useful anti-itch medications coming from this research field. |
