Efficacy of measles, mumps and rubella revaccination in children with juvenile idiopathic arthritis treated with methotrexate and etanercept

Borte S et al. – Findings suggest that low-dose methotrexate (MTX) and etanercept treatment do not seem to interfere with intended outcome of measles, mumps and rubella (MMR) revaccination in children with juvenile idiopathic arthritis (JIA).
Methods- Aim was to evaluate the influence of low-dose MTX and etanercept treatment on efficacy of MMR revaccination in children with JIA
- Markers of MMR revaccination-induced humoral and cell-mediated immunity in 15 pts (ages 6–17 yrs) were investigated
- Control group: 22 healthy children
- Production of IFN-γ by T memory cells upon in vitro stimulation with MMR antigens and seroprevalence of virus-specific IgG Abs were assessed
- Medication use, disease activity and pts' comments on side-effects were observed during 6 mo before and after revaccination
- Low-dose MTX therapy following MMR did not hamper T-cell mediated immunity
- Revaccination did not interfere with generation of long-lived virus-restricted T cells and protective levels of virus-specific IgG Abs
- No increase in disease activity or medication use seen within 6 mo after MMR revaccination, including JIA pts using etanercept
- No overt measles, mumps, rubella or secondary severe infections were noted
