Elevated synovial expression of triggering receptor expressed on myeloid cells-1 (TREM-1) in patients with septic arthritis or rheumatoid arthritis

Collins CE et al. – Synovial TREM-1 expression is elevated in septic arthritis and rheumatoid arthritis (RA); targeting TREM-1 may have therapeutic benefits by reducing local proinflammatory cytokine and chemokine release.
Methods- Aim was to determine whether synovial expression of TREM-1 is up-regulated in pts with distinct types of inflammatory or non-inflammatory arthritis
- Synovial fluid (SF) analyzed for soluble TREM-1 (sTREM-1), TNFα and leukocyte TREM 1 mRNA
- Synovial tissue from 4 RA pts, 2 pts with Crohn’s associated arthritis, 1 with AS, and 1 with OA examined for TREM-1 expression by IHC
- 3 of the RA samples also analyzed by WB
- SF sTREM-1 levels in septic arthritis and RA were similar to each other; greater than gouty arthritis, non-septic/non-RA inflammatory arthritis, and non-inflammatory arthritis
- SF TNFα and sTREM-1 levels correlated with each other
- sTREM-1 and leukocyte TREM-1 mRNA levels each correlated with SF leukocyte counts
- TREM-1 in RA was expressed in situ in synovial tissue by cells of myelomonocytic lineage; not detectable in control
