Palladini G et al. - Treatment with melphalan, thalidomide, and reduced intensity dexamethasone (MTD) is feasible in patients with advanced cardiac AL amyloidosis and effective in subjects with preserved systolic function.
Methods- Aim was to see if pts with advanced cardiac AL can tolerate aggressive therapy
- 22 pts were treated with combining oral melphalan, thalidomide, and reduced intensity dexamethasone (MTD)
- 6 pts died due to cardiac amyloidosis before completing cycle 3
- Early death was associated with reduced ejection fraction
- 8 pts achieved a hematological response and 4 a durable improvement of cardiac dysfunction
