Radioiodinated benzamide analogues have been shown to bind melanin content for targeted imaging and therapy of melanoma metastasis in the clinic. We utilized Fluorine-18 to label Dialkylamino-alkyl-benzamide via a one-step conjugation reaction. The synthesized 18F-N-[2-(diethylamino)ethyl]-4-fluoro-Benzamide (18F-FBZA) could bind to melanin contents in both melanotic melanoma cells (B16F10) and tumors. Murine-lungs bearing melanotic B16F10 pulmonary metastases displayed a significantly higher lung uptake compared to the normal lungs ( P < 0.01). 18F-FBZA based µPET and PET-CT imaging is more promising compared with 18 F-FDG PET in terms of identifications of melanotic lesions in pulmonary metastases of murine melanotic melanoma models. Thus 18F-FBZA specifically targets primary and metastatic melanotic-melanoma lesions with high tumor uptake and have great translational potentials.
