Ren G et al. – This proof–of–concept research clearly demonstrated that synthetic 2–helix 68Ga–DOTA–MUT–DS is a promising PET probe for imaging HER2 expression in vivo. The Affibody–derived small 2–helix protein scaffold has great potential for developing targeting agents for a variety of tumor–associated biomarkers.

Combining small size, simple well defined structure, high binding affinity and specificity together, Affibody molecules represent a group of nonimmunologic small protein binders which can target different tumor biomarkers. A 2-helix HER2 targeting Affibody molecule DUT-DS was chemically synthesized, conjugated with DOTA and radiolabeled with 68Ga for PET imaging of HER2 expression in mice. This novel PET probe has shown great promise in its capability of evaluations of HER2 expression and modulation in vivo. The approach could potentially benefit patients with HER2 over-expression and HER2 targeted therapy in clinic.
