BMS747158-02 is an 18F labeled agent being developed for myocardial perfusion imaging (MPI) with positron emission tomography (PET). Cardiac imaging with this agent shows clear myocardium and identification of the perfusion deficit area in rats, rabbits, pigs, nonhuman primates and human subjects. In isolated heart preparation, myocardial extraction of this agent is higher and correlates better with flow at a wide range of flow rates than currently available SPECT MPI agents. In a recent pig study, the myocardial perfusion measured by PET imaging with BMS747158-02 correlated well with flow quantified by microsphere technique both at rest and stress conditions. These findings suggest that BMS747158-02 possesses an improved chemical profile with less “roll-off” phenomena observed with the current SPECT MPI agents. In this study, impacts of feeding state and anesthetic on cardiac imaging and uptake of this agent were examined in rats in comparison with 18F-fluorodeoxyglucose (FDG). Results showed heart uptake of FDG was low and heart was not visible when imaged with FDG in rats fasted or anesthetized with ketamine and xylazine. However, unlike FDG, heart upatke of BMS747158-02 was high under these conditions. Cardiac images of this agent are clear and not affected by the feeding state and anesthetics.
