Hasturk S et al. – Phase II trial reports that the combination of gemcitabine-cisplatin-vinorelbine (GCV) with lower doses of cisplatin and gemcitabine was effective and active in advanced nonsmall cell lung cancer (NSCLC). The overall response rate, 1-year survival and median survival time were similar to higher doses; toxicities were more acceptable and manageable.
Methods- Aim was to investigate if combination of GCV with lower than usual but acceptable doses of G and C can obtain higher response rate than doublet but less toxicity than triplet combinations
- Stage IIIB and IV chemotherapy naive NSCLC pts with measurable disease and performance status of 0 to 2 were included
- Gemcitabine and vinorelbine at the doses of 900 mg/m2 and 25 mg/m2, respectively were administered on d 1 and 8, and cisplatin at a dose of 50 mg/m2 on d1/ 21 days
- 3/39 (7.7%) pts included were complete responders
- ORR was 56.4%, MTP was 6 mo, median OS time was 12 mo, and 1-yr survival rate was 49.6%
- Grade II to III neutropenia and thrombocytopenia occurred in 24% and 30% of the pts, respectively
- Febrile neutropenia was observed in 13.5% pts, and only these pts received G-CSF
- Platelet and erythrocyte transfusions were required in 12 (32.4%) pts
- No toxic or early death was observed
