Seidel P et al. - The data suggest that DMF down-regulates cytokine secretion not only by inhibiting NF-kB but a wider range of NF-kB linked signaling proteins which may explain its potential beneficial effect in asthma.

Our data suggests that beside a direct inhibtion of active NFkappaB the major anti-inflammatory action of DMF happens up-stream of NFkappaB. Since NFkappaB is not only acting as a transcription factor but also as a general modulator of histone acetylation, thereby re-structuring the DNA formation in a way that makes it accessible for other transcription factors and/or bindign of the transcritpional protein complex, it is pausible that inhibition of NFkappaB reduces gene activity over a wider range than of gens with a direct NFkappaB binding DNA sequence. Furthermore, if MSK-1 is affected by DMF this further widens the anti-inflammatorx action of DMF to a wider range of inflammation.
