Zhang H et al. – These data indicate that the new generation of synthetic anti–Hsp90 drugs, exemplified by BIIB021 that is currently undergoing Phase II testing, may have broader application against tumors with acquired multidrug resistance or tumors located in organs protected by MDR proteins, such as the adrenal glands, brain and testis.

| BIIB021 is the first synthetic Hsp90 small molecule inhibitor that enters clinical trial. Compare to the nature Hsp90 inhibitor and its derivatives, BIIB021 is not affected by the expression of multidrug resistant proteins. This implies that the small molecule inhibitor may have a broad apectrum of antitumor activity, considering that a significant amount of cancers express MDR or its related proteins, either by nature or acquired during previous treatment. |
