Multicenter, phase II study of gemcitabine and S-1 combination chemotherapy in patients with advanced biliary tract cancer

Sasaki T et al. – Gemcitabine and S-1 combination chemotherapy has promising efficacy and tolerability in patients with advance biliary tract cancer.

• 35 patients with measureable lesions and no hx of chemotherapy or RT
• Gemciatbine was administered at 1000 mg/m2 iv on d 1 & 15 q4w
• S-1 was administered po at 40 mg/m2 bid on d1-14

• 14 patients had gallbladder cancer, 14 patients had intrahepatic cholangiocarcinoma, and 7 patients had prior surgical resections
• ORR was 34.3%
• Overall disease control rate 82.9%
• Median OS 11.6 mos
• Median TTP 5.9 mos
• Grade 3/4 leukopenia (23%), neutropenia (34%), anemia (20%), thrombocytopenia (6%), and anorexia (3%)
