Review of pharmacokinetics/pharmacodynamics, in vitro and in vivo study findings, adverse effects, and therapeutic use for dasatinib
Review of all clinical studies and case reports in MEDLINE, EMBASE, and International Pharmaceutical Abstracts

Results

Observed mutations in BCR-ABL amino acid sequence cause treatment failure with existing TK inhibitors
Dasatinib has in vitro and in vivo activity against BCR-ABL, including mutations resistant to other TK inhibitors
Dasatanib had the greatest benefit in chronic-phase CML pts, with complete hematologic responses in 90% of pts
In START-R trial of dasatinib vs high-dose imatinib (800 mg/d), both regimens had comparable ability to induce a complete hematolog- ic response; more pts achieved a major cytogenetic response with dasatinib
Adverse effects: significant myelosuppression
Dasatinib has potential use in management of nonleukemic malignancies
