Methods- Review of pharmacokinetics/pharmacodynamics, in vitro and in vivo study findings, adverse effects, and therapeutic use for dasatinib
- Review of all clinical studies and case reports in MEDLINE, EMBASE, and International Pharmaceutical Abstracts
- Observed mutations in BCR-ABL amino acid sequence cause treatment failure with existing TK inhibitors
- Dasatinib has in vitro and in vivo activity against BCR-ABL, including mutations resistant to other TK inhibitors
- Dasatanib had the greatest benefit in chronic-phase CML pts, with complete hematologic responses in 90% of pts
- In START-R trial of dasatinib vs high-dose imatinib (800 mg/d), both regimens had comparable ability to induce a complete hematolog- ic response; more pts achieved a major cytogenetic response with dasatinib
- Adverse effects: significant myelosuppression
- Dasatinib has potential use in management of nonleukemic malignancies
