Methods- Human nucleus pulposus (NP) tissue was obtained post-mortem from patients without back pain and graded as normal or degenerate
- Symptomatic degenerate NP samples were also obtained at surgery for LBP
- MMP-10 mRNA and protein expression was determined by PCR and immunochemistry
- Gene expression for IL-1, TNF-alpha, NGF, and Substance P was determined
- MMP-10 mRNA was increased in surgical degenerate NP samples compared with normal and degenerate post-mortem samples
- MMP-10 protein was increased in degenerate surgical NP samples compared to normal post-mortem samples
- IL-1 and MMP-10 mRNA, but not TNF-alpha, had a significant correlation between surgical degenerate samples
- NGF was significantly correlated with MMP-10 and Substance P mRNA in surgical degenerate NP samples
