Efficacy and safety of diacerein in early knee osteoarthritis: A randomized placebo-controlled trial

Brahmachari B et al. – In patients from India with knee osteoarthritis (OA), diacerein effectively reduced pain and improved physical function. Despite frequent adverse events, overall tolerability seemed to be good.
Methods- Study evaluates the efficacy and safety of diacerein in early, symptomatic knee OA in Indian population
- 64 pts randomized to receive either diacerein or placebo for 8 wks, followed by 4 wks “treatment-free” f/u
- Primary efficacy variable: VAS assessment of pain on movement
- Secondary efficacy variables: WOMAC subscores for stiffness and physical function, rescue medication use and physician's clinical global impression (CGI)
- Compared to placebo, diacerein showed:
- Significant reductions in VAS pain scores
- Reductions in WOMAC physical function scores
- Lower requirement for rescue medication, and
- Better CGI grades
- Incidence of AEs were higher in diacerein arm: urine discoloration and soft stool being the most common ones
- However, most events were of mild to moderate intensity
