Autologous conditioned serum (Orthokine) is an effective treatment for knee osteoarthritis

Baltzer AWA et al. – The data demonstrate that autologous conditioned serum (ACS) injection considerably improves clinical signs and symptoms of osteoarthritis (OA). It remains to be determined whether ACS is disease-modifying, chondroprotective, or chondroregenerative.

• A controlled clinical trial to test the efficacy of ACS (orthokine), a novel injectable antiarthritic derived from the patient's own blood
• 376 pts with knee OA in a prospective, randomized, patient- and observer-blinded, placebo-controlled trial using an intention-to-treat analysis (ITT)
• The clinical effects of ACS were compared to hyaluronan (HA) and saline (placebo) after 7, 13 and 26 wks
• After 104 wks an observer-blinded f/u was carried out
• Frequency and severity of AEs were used as safety parameters

• In all treatment groups, intra-articular injections produced a reduction in symptoms and an improvement in QoL
• Effects of ACS were superior to those of HA and saline and improvements were clinically relevant; no differences between the effects of HA and saline
• Frequency of AEs was comparable in the ACS and saline groups, but higher in the HA group
