Filla MS et al. – Distinct beta 1 and alpha v beta 3 integrins signaling pathways converge to enhance CLAN formation. Beta 1–mediated CLAN formation was PI3–K–dependent while beta 3–mediated CLAN formation was CD47– and Rac1/Trio–dependent and may be regulated by thrombospondin–1. Both integrin pathways were Src–dependent.

CLAN formation in HTM cells has been observed in response to treatment with glucocorticoids such as dexamethasone, and it has been suggested that these actin structures may be associated with steroid-induced glaucoma (SIG). In an earlier report (Filla, et al. Invest. Ophthalmol. Vis. Sci. 2006 47:1956-67) we demonstrated integrin-dependent CLAN formation in HTM cells. In this study we sought to characterize the signaling pathways that regulate integrin-dependent CLAN formation, in part, to identify potential targets for future therapies designed to treat SIG.
