Elevated cerebrospinal fluid and plasma homocysteine levels in ALS

Valentino F et al. – Homocysteine (HC) is a biochemical marker in amyotrophic lateral sclerosis (ALS) and may be related to the disease pathophysiology.
Methods- Assay the cerebrospinal fluid (CSF) and plasma levels of HC in ALS pts and controls
- Evaluation of relationship between HC levels and clinical disease variables
- CSF from 69 (M/F 1.87) and plasma from 65 ALS pts (M/F 1.83)
- Storage at -80 degrees C until use
- Controls (55 CSF; 67 plasma): pts admitted to hospital for neurological disorders with no known relationship to HC changes
- CSF and plasma from ALS pts and controls obtained as a key step of diagnostic workup
- HC levels in CSF and plasma assayed by high performance liquid chromatograph (HPLC) and a fluorimeter detector
- Median level of total HC in CSF of ALS pts: 0.46 microM, significantly higher vs controls (0.24 microM, +91.6%)
- Similar trend for HC assayed in plasma (ALS, 12.4 microM vs controls, 7.26 microM, +70.8%)
- CSF and plasma HC levels showed no relationship with disease progression, onset age, and onset site
