Nature of "Tau" immunoreactivity in normal myonuclei and inclusion body myositis

Salajegheh M et al. – Sarcoplasmic accumulation of phosphorylated–tau has been widely stated to occur in and contribute to the pathogenesis of muscle disease in inclusion body myositis. Normal myonuclei contain neurofilament H or other unidentified 200 kDa proteins with similar phosphorylated motifs accounting for SMI–31 immunoreactivity.

| While phosphorylated-tau has been widely stated to occur in and contribute to the pathogenesis of muscle disease in inclusion body myositis, we were able to show that normal and IBM myonuclei demonstrate immunoreactivity against some of the same antibodies used for identifying "abnormally accumulated" tau by immunohistochemistry. In addition, western blots prepared from normal myonuclei (obtained through differential solubilization and laser capture microdissection) demonstrated that two of these antibodies recognize proteins other than tau, including what appears to be neurofilament H or other unidentified 200 kDa proteins with similar phosphorylated motifs. |
