Antibodies against interferon-beta in multiple sclerosis

Aarskog NK et al. – Real-time polymerase chain reaction (PCR) offers several advantages for measuring neutralizing antibody (NAB) vs the myxovirus-resistant protein A (MxA) protein induction enzyme-linked immunosorbent assay and should be used routinely for NAB among pts with multiple sclerosis (MS) using interferon-beta (IFN-beta) treatment.
Methods- Measurement of the frequency of binding antibody (BAB) and NAB against IFN-beta in sera from 827 relapsing-remitting MS (RRMS) c using various IFN-beta medications
- MxA protein induction assay (ELISA) and MxA messenger RNA (mRNA) induction assay (real-time PCR) for analysis of 137 BAB-positive sera for BAB
- Test of all 363 sera by real-time PCR
- Of the 827 pts, 363 (43.9%) had serum BAB after >12 mo of IFN-beta treatment
- Similar results for each serum in both NAB assays, indicating good correlation between MxA transcription and translation
- NAB present in 70.8% of BAB-positive sera, or 31.1% of the 827 pts with RRMS
