Glypican 5 is an interferon-beta response gene: A replication study
Cenit MDC et al. – Results confirm an association of polymorphisms within GPC5 with response to interferon-beta therapy in pts with multiple sclerosis (MS). Methods- Study to replicate results from 2 loci most strongly implicated in response to interferon-beta treatment, HAPLN1 and GPC5
- Case–control study, analyzing 199 MS pts treated with interferon-beta for ≥2 yrs and ≥2 documented relapses over 2 yrs, previous to treatment onset
- Assessment of 3 single-nucleotide polymorphisms (SNPs) in GPC5 locus and 3 SNPs in HAPLN1 locus by TaqMan technology
- Comparison of allelic frequencies between responders and nonresponders by chi-square test
Results- Responders had neither relapses nor increase in expanded disability status scale (EDSS) over 2-yr follow-up period
- Nonresponders had ≥2 relapses or an increase in EDSS of ≥1 point
- Association between GPC5 polymorphisms and response to interferon-beta therapy in MS pts, in agreement with earlier data
- The other locus studied (HAPLN1) did not show association with treatment response to interferon-beta
