Merseburger AS et al. – Activation of the PI3K pathway is significantly associated with adverse clinical outcome in renal cell carcinoma (RCC); analysis of biomarker combinations may identify high-risk pts and the need to modify treatment

Methods

  • Study of expression patterns of Akt pathway parameters PI3K, phosphatase and tensin homolog (PTEN), phosphor-Akt (p-Akt) and their combination, for prognostic value in RCC
  • Immunohistochemical analysis of tumor samples from 176 RCC pts for PTEN, p-Akt and PI3K expression
  • Correlation of expression levels to clinical variables and postoperative outcome by uniivariate and multivariate analysis

Results
  • Expression levels were independent of histologic grade and tumor stage, due to different activation levels of the PI3K/p-Akt pathway
  • PI3K protein activation was significantly associated with reduced survival times
  • Decreased long-term survival correlated with PTEN low/p-Akt high expression
