| In the midst of the excitement of pandemic influenza A (H1N1) 2009, it is important to develop and validate rapid, sensitive and accurate methods for detection of this novel virus. In collaboration between the National Microbiology Laboratory (NML) and the Capital District Health Authority (CDHA), primers designed to target the hemagglutinin of pandemic (H1N1) 2009 were combined to a pan-influenza A primer pair. The resulting duplex RT-PCR enabled simultaneous detection and confirmation of pandemic influenza A. While real-time RT-PCR methods could permit a more timely diagnosis, not all laboratories have the infrastructure to offer such testing. Our highly sensitive and specific duplex RT-PCR is undoubtedly an attractive option for laboratories without real-time RT-PCR capability. This assay uses conventional RT-PCR and generic reagents (without probes or specialized kits), thereby facilitating implementation into any molecular laboratory. Using generic reagents, supplies may also be more readily accessible as global demand for testing peaks. |
