Monsen T et al. - Based on the results from the present study the authors propose a protocol for sonication and recovery of bacteria associated with biofilm on infected implants prior to conventional culture. From the present protocol they recommend sonication for 7 minutes at 22°C at maximum effect which permit survival of Gram-negative bacteria.

| Sonication of implants is a promising method for diagnosis of prosthetic infections. In the present study we evaluate the effect of ultrasound on bacteria during different conditions during sonication as temperature, duration and composition of sonication tubes. We found that four factors were important for the inhibitory effect of sonication as; the type of micro organism, temperature of the sonication buffer, time of exposure to ultrasound (minutes) and the material and composition of sonication tube. From our results we also propose a protocol for sonication of implants which is associated with good recovery and a low a risk for contamination. The presented results is independent of the volume of sonication buffer used which is a benefit as the size of implants and the volume of sonication buffer varies considerably. The recovered numbers of bacteria are presented as CFU per implant. In clinical practice we strongly recommend all users to calibrate their sonication apparatus in order to optimize the recovery of bacteria since the effect and outcome may vary considerably between sonication units. |
