Balakumaran BS et al. – MYC binds to regulatory regions of the 4EBP1 gene. Suppression of 4EBP1 expression resulted in resensitization of MYC–expressing PrEC to rapamycin and increased autophagy. Taken together, the findings suggest that MYC expression abrogates sensitivity to rapamycin through increased expression of 4EBP1 and reduced autophagy.

Given that the MYC gene is amplified in 20-30% of advanced prostate cancers, our observation may suggest another mechanism underlying the poor clinical response observed in patients treated with single agent rapamycin analogues. We found that a dual PI3K/mTOR inhibitor was able to overcome MYC's impact and combined strategies may be more successful.
