Efficacy and safety of trabectedin in patients with advanced or metastatic liposarcoma or leiomyosarcoma after failure of prior anthracyclines and ifosfamide: results of a randomized phase II study of two different schedules

Demetri GD et al. – Superior disease control with trabectedin was demonstrated in liposarcomas and leiomyosarcomas.

• Phase II, open-label, multicenter, randomized study in patients with unresectable/metastatic liposarcoma or leiomyosarcoma after failure of prior conventional chemotherapy, including anthracylines and ifosfamide
• 270 patients were randomly assigned to one of two trabectedin regimens (1.5mg/m2 q3w vs. 0.58 mg/m2 qw x 3w)

• Median TTP was 3.7 mos vs. 2.3 mos, respectively
• Median PFS was 3.3 mos vs. 2.3 mos, respectively
• Median OS was 13.9 mos vs. 11.8 mos, respectively
• No cumulative toxicities
