Phase III trial of observation versus six courses of paclitaxel in patients with advanced epithelial ovarian cancer in complete response after six courses of paclitaxel/platinum-based chemotherapy: final results of the after-6 protocol 1

Pecorelli S et al. – Consolidation tx with 6 cycles of paclitaxel does not prolong PFS or OS in patients in CR after 1st-line paclitaxel/platinum-based regimens

Methods

200 patients with stages IIb to IV disease in clinical or pathologic CR after 6 courses of paclitaxel/platinum-based chemotherapy were randomly allocated to control or 6 courses of paclitaxel (175 mg/m2 q3w)

Results

Grade 2 or greater motor neurotoxicity and sensory neurotoxicity were reported in 11.3% and 28.0% of the paclitaxel-txd patients, respectively
After a median f/u of 43.5 mos, 107 patients had relapse, and 48 patients died
2-y PFS rates were 54% and 59% in the control and maintenance arms, respectively
2-y OS rates were 90% and 87% in the control and maintenance arms, respectively
Residual disease after initial surgery and stage were independent prognostic factors for PFS, whereas the tx arm had no prognostic relevance
