Romagne F et al. - The data show that 1-7F9 confers specific, stable blockade of KIR, boosting NK-mediated killing of HLA-matched AML blasts in vitro and in vivo, providing a preclinical basis for initiating phase 1 clinical trials with this candidate therapeutic antibody.

We describe the first therapeutic-candidate antibody (1-7F9) that specifically targets Killer Ig-like Receptors (KIR) on NK cells. Previously, results of stem-cell transplantation have implicated KIR as important determinants of tumor elimination by NK cells. When there was no signaling via KIR, the survival of patients with acute myeloid leukemia (AML) was markedly improved. The hope is that the 1-7F9 antibody may result in similar anti-tumor efficacy in patients with AML or Multiple Myeloma. Clinical trials are now ongoing.
