Vitamin K2 inhibits the growth of hepatocellular carcinoma via decrease of des-gamma-carboxy prothrombin

Cui S-X et al. – Vitamin K2 may suppress growth and invasion of hepatocellular carcinoma (HCC) cells via decrease of des-gamma-carboxy prothrombin (DCP).

Methods

Study of the efficacy of vitamin K2 on DCP production and tumor cell growth and invasion
3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay for cell growth and viability
Nude mice bearing HCC cels for in vivo efficacy of vitamin K2
A 24-well transwell chamber for HCC cell motility and invasive ability
Electrochemiluminescence immunoassay method for DCP levels in supernatant of cultures and in serum of mice
Western blot and immunohistochemical analysis for DCP expression in HCC

Results

Vitamin K2 (2-40 µM) significantly decreased levels of DCP production in supernatant of PLC/PRF/5 and HepG2 cells and in serum of nude mice bearing HCC xenografts
DCP inhibition noted on assays of Western blot analysis in HCC cultures and immunohistochemical analysis in HCC xenografts in mice
Vitamin K2, administration inhibited HCC growth capacity and decreased tumor cell invasion and migration
Inhibitory effects of HCC growth noted in vivo
Sensitivity well correlated with DCP decrease in serum of mice
