Cemeus C et al. – Lovastatin may augment cytotoxic effects of gefitinib and provide a rationale for combined statin/ epidermal growth factor receptor (EGFR) targeted therapies in glioblastoma

Methods

  • Study to identify agents that enhance anti-cancer effects and promote responsiveness to EGFR kinase inhibitors in glioblastoma
  • Isogenic model system of U87MG glioblastoma cells expressing EGFRvIII and PTEN in relevant combinations

Results
  • Targeting HMG-CoA reductase with lovastatin significantly enhanced glioblastoma cell sensitivity to the EGFR kinase inhibitor gefitinib in 5 cell lines
  • Combined gefitinib and lovastatin treatments induce potent synergistic cytotoxicity irrespective of EGFRvIII and PTEN status
