Ghadersohi A et al. – An inverse correlation is reported between prostate-derived Ets transcription factor (PDEF) and survivin expression, suggesting increased PDEF expression with reduced survivin is associated with prolonged survival in ovarian cancer pts

Methods

  • Study of PDEF expression in tumors of ovarian cancer pts and its association with survival
  • Evaluation of samples from 40 ovarian cancer pts

Results
  • Higher levels of PDEF mRNA and protein and lower levels of survivin in early-stage (IA) and borderline (IIB, III) ovarian tumors vs late-stage ovarian tumors (IIIC and IV)
  • Highest levels of PDEF mRNA and protein in normal ovarian vs tumor tissues
  • Overall survival significantly longer for pts with PDEF-positive and survivin-negative ovarian tumors vs PDEF-negative and survivin-positive tumors
  • Forced PDEF expression in PDEF-negative ovarian tumor cells inhibited tumor cell growth, induced apoptosis, and downregulated survivin expression and its promoter activity
  • Treatment of ovarian cancer cells with vitamin D or a selenium compound resulted in PDEF re-expression, survivin downregulation, apoptosis induction, and tumor cell growth inhibition, vs untreated controls
