McWilliams RR et al. – Minor alleles in DNA repair genes XPF/ERCC4 and XPD/ERCC2 are associated with altered risk for pancreatic cancer

Methods

  • Clinic-based, case-control study of 481 consecutive pts with confirmed pancreatic adenocarcinoma and 625 healthy controls
  • Comparison of allele and genotype frequencies for 16 single-nucleotide polymorphisms (SNPs) in DNA repair genes ERCC1, XPD/ERCC2, XPC, XPF/ERCC4, OGG1, and XRCC1 after adjusting for age, sex, smoking history
  • Subgroup analysis by sex and smoking history

Results
  • Pancreatic adenocarcinoma risk decreased in carriers of 1 or 2 XPF/ERCC4 minor alleles at R415Q vs carriers of 2 major alleles
  • Heavy smokers (>40 PY) had increased cancer risk if they carried >=1 minor allele for XPD/ERCC2 at D312N or D711D
  • No other significant differences in risk identified
