Liu Z-F et al. – A modified FOLFOX-4 regimen (biweekly oxaliplatin [OXA] with continuous infusional 5-fluorouracil [5-FU] and leucovorin [LV] every 2 wks) is active and well-tolerated in elderly pts with advanced gastric cancer (AGC); OXA may cause mild hypersensitivity

Methods

  • Study of toxicity and clinical activity of modified FOLFOX-4 regimen in 44 previously untreated AGC pts age ≥ 65
  • Administration of OXA 85 mg m-2 on day 1, LV 200 mg m-2 as 2-hr infusion followed by a 22-hr infusion of 5-FU 1000 mg m-2, repeated for 2 consecutive days every 2 wks

Results
  • All pts assessable for toxicity and 40 pts for response; median age 69 yrs
  • Response rate by RECIST criteria: 52.5%: 3 complete responses, 18 partial responses, 11 stable diseases, and 8 progressions
  • Median time to progression: 6.5 mo; median overall survival: 10 mo
  • Grade 3 hematologic toxicities: neutropenia (6.8%); anemia (2.3%); thrombocytopenia (4.5%); no grade 4 toxicities
  • Peripheral neuropathy: grade 1 in 34.1%; grade 3 in 2.3%
  • Acute hypersensitivity reaction in 1 pt
