Liang R et al. – Induction regimen of cytosine arabinoside, daunorubicin, and etoposide had acceptable toxicity, with efficacy comparable with that of standard regimens

Methods

  • Study of induction regimen in 46 pts with previously untreated acute myeloid leukemia
  • Administration of cytosine arabinoside (100 mg/m2/day as 18-hr IV infusion for 7 days), daunorubicin (50 mg/m2/day as IV bolus injection for 3 days), and etoposide (75 mg/m2/day by 1-h IV infusion for 7 days)

Results
  • Complete remission for 30 pts (67%); further consolidation and maintenance chemotherapy administered
  • Of 31 complete responders, 15 (48%) relapsed
  • Median disease-free survival of 31 complete responders: 25 mo
  • Median overall survival of all 46 pts: 14 mo
  • None of the clinical characteristics (sex, age, FAB morphology, extramedullary disease, initial WBC count) predicted clinical response
  • Toxicity: major toxicity was myelosuppression; acceptable non-hematologic side effects
