Interleukin-4 cytotoxin therapy synergizes with gemcitabine in a mouse model of pancreatic ductal adenocarcinoma

Shimamura, T., et al. – Combination IL-4 cytotoxin with gemcitabine may be effective therapy for treatment of pancreatic ductal adenocarcinoma (PDA).

Methods

  • Metastatic and orthotopic mouse model using green fluorescent protein–transfected cancer cells and whole-body imaging system to confirm synergistic antitumor activity in vivo and monitor real-time disease progression

Results
  • Moderate- to high-density surface IL-4 receptor (IL-4R) was seen in 60% of tumor samples from PDA pts versus no or low-density IL-4R in normal pancreatic samples
  • IL-4 cytotoxin was specifically and highly cytotoxic to 6 of 8 pancreatic cancer cell lines
  • Combination IL-4 cytotoxin with gemcitabine had synergistic antitumor activity in vitro
  • Combined agents completely eradicated tumors in 40% of nude mice with small established PDA tumors
  • Combined treatment significantly prolonged survival of nude mice with advanced distant metastatic PDA tumors on day 14
