Visser PJ et al. – Alzheimer's disease (AD) is a common cause of subjective cognitive impairment (SCI), non-amnestic mild CI (naMCI), and amnestic mild CI (aMCI) and is associated with cognitive decline in pts with naMCI or aMCI. Pts with SCI may be in early-stage AD, and cognitive decline might become apparent only after longer follow-up.
Methods- Study of the prevalence of a CSF AD profile in pts with SCI, naMCI, or aMCI
- Investigation of the association of this profile with cognitive outcome in each group
- Prospective cohort study of pts with SCI, naMCI, aMCI, and neurologically healthy controls from 20 European memory clinics January 2003-June 2005
- CSF AD profile defined as an abnormal ratio of Abeta42:tau
- Annual assessment of pts up to 3 yrs
- Outcome measures: changes in memory, overall cognition, Mini-Mental State Examination (MMSE) score, daily function, and progression to AD-type dementia
- CSF AD profile more common in pts with SCI (31/60 [52%]), naMCI (25/37 [68%]), and aMCI (56/71 [79%]) vs healthy controls (28 of 89 [31%])
- Profile associated with cognitive decline in naMCI pts (memory, MMSE, daily function) and in aMCI pts (MMSE, daily function)
- In aMCI pts, CSF AD profile predictive of AD-type dementia
