Poorly differentiated thyroid carcinomas (PDTC) are very aggressive tumors for which there is, presently, no effective form of treatment. The aim of our work was to identify molecular mechanisms involved in thyroid tumor progression. By microarray analysis, we have highlighted several mechanisms that are deregulated in PDTC, such as cell proliferation, poor prognosis, spindle assembly checkpoint and cell adhesion. In particular, PDTC had over-expression of UHRF1, which has an essential role in cell cycle progression and in the maintenance of DNA methylation, and under-expression of ITIH5, which encodes one of the heavy chains of the inter-?-trypsin inhibitor family, a group of proteins that play a role in extracellular matrix stabilization and in prevention of tumor metastases. Therefore, these two genes may be important in the design of novel therapeutic targets for the poorly differentiated forms of thyroid cancer.
