Robinson–White AJ et al. – 8–Cl–ADO in addition to unambiguously inhibiting proliferation and inducing apoptosis in a PKA–independent manner also has PKA–dependent effects that are unmasked by a mutant PRKAR1A. Thus, 8–Cl–ADO could serve as a therapeutic agent in patients with Carney complex–related tumors.

This work presents a mechanism of action for the drug 8-Cl-Adenosine and may resolve the ongoing controversy as to its mechanism of action on cancer cell proliferation. 8-Cl-Adenosine may be an effective drug for the treatment of Carney complex and protein kinase-A related tumors, and may act on other cancers as well.
