Association of PDE11A global haplotype with major depression and antidepressant drug response

Luo H-R et al. – Findings suggest that the cyclic nucleotide phosphodiesterase gene 11A (PDE11A) global haplotype is associated with both major depressive disorder (MDD) and antidepressant drug response.
Methods- Study of whether the whole region 2q31–q32 contributes to MDD or antidepressant response
- Review of 278 depressed Mexican-American pts and 321 matched healthy controls
- Analysis of global haplotype by examining 16 single-nucleotide polymorphisms (SNPs) in PDE11A and 6 SNPs in PDE1A
- Combination genotype at rs1880916 (AG/AA) and rs1549870 (GG)
- None of the haplotypes consisting of 6 SNPs in PDE1A show significant difference between depressed vs control groups
- Of haplotypes consisting of 16 SNPs across 440 kb in PDE11A gene, 18 common haplotypes (with frequency >0.8%) noted in studied population
- Significantly different frequencies between MDD group vs controls for 6 haplotypes
- Phylogenetic network result for the 16 SNPs showed several historic recombination events in PDE11A gene
- Significantly lower frequency of 1 haplotype in remitter vs nonremitter group for depressed pts treated with either desipramine or fluoxetine
